Udenafil was developed by the pharma company called Dong-A Pharmaceutical. Description

Udenafil is a new phosphodiesterase type 5 (PDE5) inhibitor used to treat erectile dysfunction (ED). It has been approved in South Korea for treatment of erectile dysfunction in November 2005 under the name Zydena. It is not yet approved for use in the U.S., E.U., or Canada.

Mechanism Of Action

Udenafil inhibits the cGMP specific phosphodiesterase type 5 (PDE5) which is responsible for degeneration of cGMP in the corpus cavernosum located around the penis. Penile erection during sexual stimulation is produced by increased penile blood flow caused by the relaxation of penile arteries and corpus cavernosal smooth muscle. This reaction is mediated by the release of nitric oxide (NO) from nerve terminals and endothelial cells, which stimulates the synthesis of cGMP in smooth muscle cells. Cyclic GMP causes smooth muscle relaxation and increased blood flow into the corpus cavernosum. The inhibition of phosphodiesterase type 5 (PDE5) by udenafil reinforces erection by increasing the amount of cGMP.

The amount of time Udenafil takes to work varies from person to person, but it normally takes between half an hour and one hour. However, Udenafil may take longer to start working if you take it with a heavy meal. Its effect may last for about 3-4 hours.

Udenafil reached maximum plasma concentrations at 0.8–1.3 h, and then declined mono-exponentially with a terminal half-life of 7.3–12.1 h in the single-dose study. The area under the time–concentration curves (AUC) and maximum plasma concentrations (Cmax) increased supraproportionally with increasing dosage in the single-dose study. During multiple dosing of udenafil, a stable state was reached at 5 days and little accumulation occurred after repeated dosing for 7 days.
